چکیده
G-protein coupled estrogen receptor (GPER) is a transmembrane that mediates non-genomic effects of estrogen. This study aimed to investigate the role GPER in stemness formation and malignancies triple negative breast cancer (TNBC) cells. Spheroids MDA-MB-468 cells were induced by mammosphere culture, proportion CD44+ /CD24−/low stem cell subpopulation was detected. Malignant characteristics, expression stemness-related markers, tumorigenesis xenograft assay compared between mammospheres adherent cultured The impacts 17β-estradiol (E2) GPER-specific antagonist G15 studied vitro assays. increased cells, which also showed higher markers than abilities spherical colonies proliferate, invade, form soft agar enhanced. Spherical exhibited stronger ability assay. E2 treatment enhanced tumorigenicity both treated with had proliferation, invasion, colony other groups. Pretreatment effectively blocked stimulation E2. In conclusion, TNBC positively related malignant features.

G protein-coupled KISS1 receptor is overexpressed in triple negative breast cancer and promotes drug resistance
Triple-negative breast cancer (TNBC) lacks the expression of estrogen receptor α, progesterone receptor and human epidermal growth factor receptor 2 (HER2). TNBC patients lack targeted therapies, as they fail to respond to endocrine and anti-HER2 therapy. Prognosis for this aggressive cancer subtype is poor and survival is limited due to the development of resistance to available chemotherapies...
متن کاملG-protein Coupled Receptor Dimerization
A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...
متن کاملG-protein-coupled receptor 81 promotes a malignant phenotype in breast cancer through angiogenic factor secretion
G-protein-coupled receptor 81 (GPR81) functions as a receptor for lactate and plays an important role in the regulation of anti-lipolytic effects in adipocytes. However, to data, a role for GPR81 in the tumor microenvironment has not been clearly defined. Here, GPR81 expression in breast cancer patients and several breast cancer cell lines was significantly increased compared with normal mammar...
متن کاملIdentity of an estrogen membrane receptor coupled to a G protein in human breast cancer cells.
Although nonclassical estrogen actions initiated at the cell surface have been described in many tissues, the identities of the membrane estrogen receptors (mERs) mediating these actions remain unclear. Here we show that GPR30, an orphan receptor unrelated to nuclear estrogen receptors, has all the binding and signaling characteristics of a mER. A high-affinity (dissociation constant 2.7 nm), l...
متن کاملEstrogen Receptor Mediates the Radiosensitivity of Triple-Negative Breast Cancer Cells
BACKGROUND This study aimed to evaluate differences in the radiosensitivities of triple-negative breast cancer (TNBC) and luminal-type breast cancer cells and to investigate the effects of estrogen receptor (ER) expression on the biological behaviors of the cells. MATERIAL AND METHODS Colony-forming assays were performed to detect differences in radiosensitivities in breast cancer cell lines. G...
